Selection of Primers in eDNA Metabarcoding Affects Soil Invertebrate Diversity Assessment—Evidence from a Comparative Survey

The eDNA metabarcoding greatly enhances soil invertebrate diversity surveys, compared to traditional methods. However, its application in environmental sample analyses requires careful planning in order to ensure comprehensive data. A critical step in planning is the choice of adequate primer pair to obtain the most accurate information on taxa present in the sample. In this study, primer pair mlCOIintF/dgHCO2198 was used in eDNA metabarcoding study of soil invertebrates from solonchak, resulting in detection of 14 invertebrate species. To evaluate the effectiveness of these primers, we compared the results with a previous study, in which the same soil samples were analyzed using primers BF2/BR2. The primer pair mlCOIintF/dgHCO2198 provided a better insight into the invertebrate community of solonchak soil type and detected a larger number of taxonomic categories compared to the primer pair BF2/BR2. This study demonstrates that the choice of primer pairs can affect the results obtained through eDNA metabarcoding.
