Cloning and Expression Studies of the Pitx1 Gene from Pampus argenteus Following siRNA Interference

In order to investigate the correlation between the Pixt1 gene and the development of the pelvic fin of Pampus argenteus (P. argenteus), we utilized PCR, qPCR and siRNA interference techniques to clone the ORF frame of Pixt1 gene, detect the difference in Pitx1 gene expression between different tissues of P. argenteus, and detect the changes of expression of Pitx1 and its upstream and downstream genes in P. argenteus after siRNA interference. The results showed that the Pitx1 gene contained an 858 bp ORF frame encoding 285 amino acids. The Pitx1 protein encoded by the Pitx1 gene contains a Hox domain. The expression of the Pitx1 gene between different tissues showed that the Pitx1 gene, a key gene for the formation of hindlimb cartilage, was low in the abdominal epithelium (primary pelvic fin growth site) of P. argenteus, showing no hindlimb specificity. Interference with the Pitx1 gene will affect the expression of its upstream and downstream genes, including the downregulation of the Tbx4 gene and the upregulation of the Fgf10 gene. These results can provide a theoretical basis for further study on the loss of pelvic fins of P. argenteus.
