Comparative Effectiveness of Different DNA Barcodes for Species Delimitation in Microalgae of the Clade Chlorella (Chlorophyta)

Comparative effectiveness of species delimitation using three popular DNA barcodes was analyzed in the microalgal clade Chlorella. The V4 and V9 regions of the 18S rRNA gene were found to have low resolution and were the least effective for species-level identification. The ITS2 marker had a higher accuracy and was recognized as the optimal choice for delimitation of species in this clade. Comparison of delimitation algorithms showed that the methods based on combining the phylogenetic and coalescent models and on comparing the number of clusters with estimates of genetic diversity had an advantage for the analysis of the studied set of sequences. The use of ITS2-based metabarcoding for communities of Chlorella microalgae is recommended to obtain reliable data on their biodiversity in various ecosystems.
