Abstract <p>Exopolysaccharides (EPSs) of microbial origin are among the most thoroughly investigated bioactive compounds due to their fascinating properties, including anticancer, anti-inflammatory, antioxidant, and antibacterial effects. The present work aimed at isolation, characterization, and assessment of the antibacterial activity of EPSs produced by strains isolated from different biotopes in Algeria. Initially, bacterial isolates were screened, and those forming mucoid colonies on an HJL agar were selected for secondary screening to evaluate EPSs production by gravimetric analysis of EPSs dry weight. As a result, a single strain designated CH1, isolated from a wastewater sample, was selected for further studies. In accordance with phylogenetic analysis of the partial sequence of the PCR-amplified and sequenced 16S rRNA gene, strain CH1 was identified as belonging to the genus <i>Agrobacterium</i> and was found to be closely related to <i>Agrobacterium radiobacter</i>, with a 100% identity in the 16S rRNA gene sequence. The strain CH1 demonstrated the maximum yields of EPS production (1.98 ± 0.7 and 1.98 ± 0.5 g, respectively) when glucose and sucrose were added to HJL agar medium at 3 and 1% (wt/vol) concentrations. FTIR analysis confirmed the presence of polysaccharide functional groups in the crude EPSs, which were further identified by <sup>1</sup>H NMR, revealing acetyl, succinyl, and pyruvyl groups. At 20 and 40 mg/mL, these EPSs exhibited inhibitory zones of 10 and 13 mm, respectively, against <i>Staphylococcus aureus</i> ATCC 25923, indicating significant antibacterial activity. Given these findings, we propose these EPSs as promising antibacterial agents for the management of <i>Staphylococcus aureus</i> infections.</p>
