Development of a Transformation Protocol for Lacticaseibacillus paracasei RG

Lactic acid bacteria, traditionally used in the food industry, are considered as a platform for development of recombinant strains capable of expressing antigens of protective significance and other biologically active molecules and delivering them into the gastrointestinal tract of farm animals and humans. One of the obstacles to obtaining recombinant strains of lactic acid bacteria is the low efficiency of their transformation with plasmid DNA. This study was carried out in Lacticaseibacillus paracasei RG, selected as a safe probiotic strain capable of crossing the acidic barrier of the stomach. We assessed how cultivation conditions, method of competent cell preparation, and electroporation mode affected the efficiency of L. paracasei RG transformation by the pTRKH2 vector. The results were used to develop a protocol for efficient transformation of L. paracasei RG with recombinant DNA.
