Gut Microbiota Composition of American Horseshoe Crabs (Limulus polyphemus) from Yucatan Peninsula

Despite the medical and ecological relevance of horseshoe crabs (a species that has managed to survive for approximately 450 million years), few studies have focused on determining the composition of microbial communities in their intestines. This study used high-throughput 16S rRNA gene sequencing for comparative analysis of the gut microbiota composition of the American horseshoe crab Limulus polyphemus at three sites (Rio Lagartos [RL], Chabihau [CHB] and Champoton [CHAMP]) from the Yucatan Peninsula, Mexico: RL and CHB are located to the north of the peninsula and CHAMP to the south-southwest. The results revealed that the microbiota of L. polyphemus was composed mainly of Pseudomonadota (RL: 67.7%, CHB: 57.1% and CHAMP: 42.2%), ‘Candidatus Parcubacteriota’ (RL: 7.8%, CHB: 8.7% and CHAMP: 0.1%), Bacillota (RL: 4.0%, CHB: 8.3% and CHAMP: 11.7%), Actinomycetota (RL: 2.9%, CHB: 16.5% and CHAMP: 12.1%), Fusobacteriota (RL: 3.1%, CHB: 0.2% and CHAMP: 12.2%) and Bacteroidota (RL: 2.3%, CHB: 2.2% and CHAMP: 7.7%). The composition of the microbiota of L. polyphemus from RL and CHB was relatively similar and differed significantly from that of CHAMP. The present study is the first report on the composition of the microbiota of L. polyphemus, helping to understand its internal biology.
