Mouse C2C12 Myoblasts as a Model for Studying Piezo1 Mechanosensitive Channels in Skeletal Muscle

Immortalized mouse C2C12 myoblasts are an important modelfor studying the physiology and pathophysiology of skeletal muscle.Since muscle tissue function is inextricably linked to mechanical processes,investigating the activity of various mechanosensory molecules andsignaling cascades is a crucial task. Mechanically gated cationchannels Piezo1 play a key role in cellular mechanotransductionand mechano-dependent responses in both non-excitable and excitablecells and tissues. Elucidating the role of Piezo1 and the mechanismsof its regulation in C2C12 cells is an important direction in myology,yet remaining in its infancy. In this review, we compile experimentalevidence on Piezo1 function and discuss possible signaling pathwaysand specific features of Piezo1 regulation in C2C12 cells. Identifyingthe patterns governing Piezo1 activity and associated cellular signalingprocesses in C2C12 cells may reveal new aspects of Piezo1 involvement thatare important not only for fundamental and applied myology but alsofor other areas of mechanobiology.
