Barrier Properties of the Intestinal Epithelium in the Dynamics of Nephropathy Progression

This study investigated the effect of 5/6 nephrectomy on thebarrier properties of the jejunum and colon of rats in the long-termpostoperative period (two, four, and six months). It was shown that animalswith kidney dysfunction developed progressive azotemia, hypernatremia,and hyperkalemia, increased levels of uremic toxins (trimethylamineand trimethylamine-N-oxide), and elevated blood pressure. Two monthsafter nephrectomy, the jejunum exhibited a decrease in short-circuitcurrent and an increase in paracellular permeability to sodium fluorescein,whereas transepithelial resistance (TER) remained unchanged. Inthe colon at the same time point, an increase in TER and permeabilitywas observed along with a decrease in short-circuit current. Bythe fourth month, the jejunum retained morphological alterationsin the villi and crypts, but the barrier function stabilized dueto a compensatory increase in occludin levels and a decrease inclaudin-1. In the colon, increased permeability persisted withoutsignificant changes in the level of the studied tight junction proteins.Six months after nephrectomy, the differences in permeability andthe content of tight junction proteins compared to the control groupwere leveled out, although morphological disorders of the epithelium persisted.The results demonstrate the potential of the intestinal epitheliumto restore intestinal barrier properties following nephrectomy.
