<p><i>Fusarium</i> and <i>Neocosmospora</i> are widely recognized genera pathogenic to humans, animals, and plants. While the resistance of clinical and environmental strains to antifungal drugs and fungicide agents has been extensively studied, there is still limited knowledge regarding the susceptibility profiles within the One Health context. This study aims to investigate the susceptibility of clinical, animal, and environmental isolates collected during a One Health project conducted in Colombia from 2021 to 2023 to four antifungals i.e. amphotericin B, voriconazole, itraconazole, and posaconazole, as well as four fungicides, i.e. imazalil, propiconazole, tebuconazole, and difenoconazole. The results revealed that most clinical, animal, and environmental isolates were susceptible to amphotericin B and voriconazole. However, all tested isolates exhibited high Minimum Inhibitory Concentrations (MIC) for itraconazole, posaconazole, propiconazole, tebuconazole, and difenoconazole. These findings underscore the importance of initiating surveillance for antifungal and fungicide susceptibility within the framework of the One Health initiative.</p>
