<p>A novel actinomycete strain, cg5<sup>T</sup>, was isolated from the rhizosphere soil of <i>Cyclosorus parasiticus</i>, collected from Xiangtan City, Hunan province, China. Strain cg5<sup>T</sup> contained <i>meso</i>-diaminopimelic acid as a diagnostic amino acid. Whole-cell sugars of strain cg5<sup>T</sup> included arabinose, galactose, ribose, and rhamnose. The chemotaxonomic properties of this strain are typical of members of the genus <i>Amycolatopsis</i>. The genomic DNA G + C content of strain cg5<sup>T</sup>, whose genome size is approximately 9.7 Mb, was 68.5%. The full-length 16S rRNA sequence analysis indicated that this strain was a member of the genus <i>Amycolatopsis</i> and shared the highest sequence similarity to <i>Amycolatopsis xylanica</i> CPCC 202699<sup>T</sup> (99.23%). Phylogenetic trees based on 16S rRNA gene and genome sequences revealed that strain cg5<sup>T</sup> was most closely related to <i>A. xylanica</i> CPCC 202699<sup>T</sup>. However, the ANIm and dDDH values between these two strains were 92.23% and 45.80%, respectively, far lower than the 96.6% ANIm and 70% dDDH cut-off points recommended for delineating species. Moreover, phenotypic and chemotaxonomic characteristics further confirm that strain cg5<sup>T</sup> represents a new species of the genus <i>Amycolatopsis</i>, for which the name <i>Amycolatopsis cyclosori</i> sp. nov. is proposed. The type strain is cg5<sup>T</sup> (=MCCC 1K09227<sup>T</sup> = KCTC 59391<sup>T</sup>).</p>
