<p>Shadow puppetry, an ancient Chinese art blending craftsmanship and opera, is historically and artistically significant. However, the long-term preservation of animal-hide puppets has led to severe deterioration, particularly microbial degradation. This study conducted airborne microbial sampling in three preservation environments (exhibition halls, showcases, and warehouses) of shadow puppet relics at the National Shadow Puppetry Museum in Chengdu. We identified the primary fungi and bacteria (<i>Bacillus subtilis</i>, <i>Staphylococcus</i>, <i>Aspergillus</i>, and <i>Penicillium</i>) and confirmed their collagen-degrading abilities, identifying them as potential threats to these relics. Further antibacterial experiments showed that 0.3% BC01 (isothiazolinone) and 40 mg/ml carvacrol had significant inhibitory effects on the main degrading microorganisms, with BC01 having a better effect on fungi such as <i>Aspergillus flavus</i>, while carvacrol had a more significant inhibitory effect on <i>Talaromyces marneffei</i> and bacteria. Hence, this study analyzed two antibacterial reagents which can be used to lower the shadow puppet preservation environment’s microbial contamination and provides critical insights for conserving leather-based cultural relics.</p>
