Comprehensive in-vitro study of anticholinesterase, anti-inflammatory, and antidiabetic properties of congress grass (Parthenium hysterophorus) methanolic extract

Parthenium hysterophorus is a weed and flowering plant belonging to the Asteraceae family. This study investigated the in vitro activity of P. hysterophorus’s aerial part. The α-amylase and α-glucosidase assays were performed to assess the anti-diabetic activity of the methanolic extract of the plant’s aerial part. In addition, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) activity assays were conducted to evaluate the anti-cholinesterase activity. Moreover, cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) activity assays were performed to explore the potential of P. hysterophorus in managing inflammation. The P. hysterophorus methanolic extract demonstrated α-amylase and α-glucosidase inhibition (IC50 36.46 and 123.88 µg/mL, respectively) comparable to that of acarbose (26.58 and 21.30 µg/mL, respectively). The methanolic extract of P. hysterophorus was a moderate inhibitor of AChE (61.44%) and BChE (64.98%) at a concentration of 1000 μg/mL. In comparison to standard drugs, i.e., celecoxib (IC50 3.22 μg/mL) and montelukast (IC50 4.50 μg/mL), the methanolic extract of P. hysterophorus also exhibited significant inhibition against COX-2 and 5-LOX showing IC50 values of 13.52 and 11.53 μg/mL, respectively. Therefore, P. hysterophorus possessed effective anti-inflammatory, anti-diabetic, and anti-cholinesterase activities. Further in vivo and clinical studies should be conducted on specific bioactive components present in this plant to validate these therapeutic properties.
