Extraction optimization and bioaccessibility of phytochemicals of a herbal mixture

To achieve the best antioxidant performance along with the highest total polyphenol content of a medicinal herbal mixture, a Face Centered Composite Design (FCCD) is performed to determine the optimized extraction parameters. Moreover, the bioaccessibility of the herbal mixture polyphenols is evaluated via an in vitro gastrointestinal digestion. The FCCD determined the maceration parameters as 73.35% of ethanol, 56.01 min of rotation and a ratio of 0.10 (dry weight/ solvent). The hydro-ethanol herbal mixture obtained following the optimized maceration process showed a total polyphenol content of 73.77 mg of GAE/ g DW and a total flavonoid content of 9.77 mg RE/ g DW. Extracts are rich in rosmarinic acid, rutin and carnosic acid and showed a good DPPH scavenging activity with an IC50 of 0.25 mg extract/ mL. The bioaccessibility of total polyphenols showed to be 61.21% and that of the total flavonoids was 48.83%. The digested fraction revealed a good DPPH scavenging activity with an IC50 of 0.40 mg extract/ mL. The current investigation highlights the potential use of rosemary-lemon verbena-carob mixture as nutraceuticals.
