<p><i>Cocculus hirsutus</i> (L.) Diels (Menispermaceae), commonly known as ‘Jal-jamni’ distributed in tropical countries. The plant has been used in India’s traditional medicinal systems for the treatment of diabetes, cancer, arthritis, inflammation, cardiac, immune diseases, fever, spermatogenesis, microbial infections, and urinary diseases as well as being used traditionally in therapeutics. These biological activities were recorded in plant due to the presence of phytochemicals, such as corsutine (1), coclaurine (2), cohirsinine (3), cohirsine (4), cohirstinine (5), cocsuline-N-2-oxide (6), cohirsitinine (7), hirsutine (8), haiderine (9), jamtinine (10), jamtine (11), mangiflorine (12), shaheenine (13), isotrilobine (14), syringaresinol (15), isoquinoline (16), rutin (17), hirsutidin 3-glucoside (18), and liquiritin (19). In addition, recently, two new alkaloids were reported from the roots of <i>C. hirsutus</i> such as 15-carboxydihydroerysotrine and (14 R)-4-methoxy-13,14-dihydrooxypalmatine. A recent Clinical Trials Registry-India comparative, multi-centre, controlled clinical study has been initiated in India to examine the consequences and safety of aqueous extract of <i>C. hirsutus</i> tablets in the mitigation of Corona virus infection. The present review focuses on the bioactive metabolites, biological activities, and traditional use of <i>C. hirsutus.</i> Plant extracts are also useful, safe, and harmless, which could contribute to their application in therapeutics. In the future, research on in vivo and clinical studies is necessary to assess the pharmacological actions of <i>C. hirsutus</i> along with metabolite synthesis pathway elucidation studies to isolate and identify the individual bioactive metabolites.</p>
