<p>Indigenous mushrooms play a vital role in enhancing nutrition, promoting food security, and supporting livelihoods in Nigeria and Kenya. Despite their ecological and socio-economic significance, there remains a lack of comprehensive reviews addressing the domestication and commercialization of these valuable species. This review was written to explore and consolidate knowledge on the domestication challenges and opportunities for indigenous mushrooms in these regions, addressing a critical gap in the existing literature. A systematic literature review was conducted using academic databases such as Scopus, PubMed, and Google Scholar, focusing on peer-reviewed articles, institutional reports, and case studies published between 2000 and 2024. Selection criteria emphasized relevance to mushroom species indigenous to sub-Saharan Africa, particularly those with documented cultivation or commercial potential. Findings reveal that while various indigenous species are traditionally utilized, domestication efforts remain limited by factors such as environmental constraints, pest and disease susceptibility, and knowledge gaps. Current cultivation methods range from wild harvesting to semi-controlled substrate-based systems, though scalability remains a challenge. Nutritional analyses underscore the richness of these mushrooms in essential micronutrients and bioactive compounds, with potential to address widespread nutritional deficiencies. Consumer preferences are shaped by cultural perceptions, taste, and health considerations, influencing regional market trends and acceptance. Innovative approaches such as the use of agricultural waste substrates, biotechnological tools (e.g., CRISPR, metabolomics), and digital farming technologies offer promising avenues for improved yield and quality. Indigenous mushrooms hold untapped potential for advancing nutrition, health, and rural economies in Nigeria and Kenya. The aim of this review is to systematically examine the cultivation techniques, nutritional benefits, consumer preferences, and market potential of indigenous mushrooms in Nigeria and Kenya, while identifying research priorities and strategies to unlock their full socio-economic value.</p>
