Pathomorphological findings and antigen distribution in goats experimentally infected with Mycoplasma mycoides subsp. capri (Mmc)

Contagious caprine pleuropneumonia, caused mainly by Mycoplasma mycoides subspecies capri (Mmc), is an inherent threat to the goat population of India. This pathogen majorly affects lung pathophysiology. The present study describes the novel pathological and immunohistochemical findings in less commonly explored organs of 5 Jamnapari goats experimentally infected with Mmc. In this study, five adult goats, approx. 1–1.5 years-age and either sex, were inoculated with Mmc @ 1 × 108 CFU dissolved in 1 ml PPLO broth via endobronchial route, while other 2 adult goats were kept as control. Clinically, the infected goats showed high fever (104–106°F), serous-to-mucopurulent nasal discharges, tachypnea, and 100% mortality within a week from inoculation. The inoculated goats showed predominant microscopic lesions of acute fibrinous necrotizing suppurative pleuropneumonia and tracheo-bronchial lymphadenitis. However, the other vital organs also showed varied microscopic lesions such as non-suppurative myocarditis, hepatitis, glomerulo-nephritis, abomasitis, enteritis, splenitis, lymphadenitis, orchitis, metritis, and meningitis. Moreover, the distribution of Mmc antigens at the lesional sites, reisolation of Mmc, along with the molecular detection confirmed the tissue pathologies due to mycoplasmaemia. High intensity immunosignals of Mmc antigens in the lymph nodes, spleen and the adrenal glands, in addition to lungs, might suggest for their incorporation in sampling plan for the diagnosis and isolation of pathogen. The results presented in the study provide novel insights into tissue tropism and pathogenesis of Mmc in goats.
