Salicylic Acid and Sodium Nitroprusside Improve the Drought Tolerance of Henna (Lawsonia inermis L.) by Modulating Its Physiological and Biochemical Characteristics

The target of the present study was to determine the effects of exogenous salicylic acid (SA) and sodium nitroprusside (SNP) on the physiological and biochemical characteristics of henna under different levels of drought stress. In the present study, we employed a split-plot experiment based on a randomized complete block design with three replicates. The main factor included three distinct levels of drought stress: 95% Field Capacity (95% FC), 75% Field Capacity (75% FC), and 50% Field Capacity (50% FC), and the sub-factor included control and three growth regulators, SA (0.25 mM), SNP (50 µM), and SA + SNP. We measured the relative water content, photosynthetic pigments, photosynthesis parameters, chlorophyll florescence, proteins, malondialdehyde, and the response of various antioxidants in henna. The results showed that the foliar application of salicylic acid + sodium nitroprusside under 50% field capacity conditions increased protein content (18.49%), chlorophyll a (39.43%), chlorophyll b (44.73%), total chlorophyll (40.55%), carotenoids (11.29%), F0 (9.29%), Fm (3.3%), Fv/Fm (1.08%), net photosynthesis rate (5.69%), transpiration rate (5.20%), leaf stomatal conductance (7.42%), catalase activity (20%), peroxidase activity (18%), polyphenol oxidase activity (18.18%), guaiacol peroxidase activity (6.95%), phenylalanine ammonia-lyase activity (23.76%), total phenol (8.30%), anthocyanin (15.67%), and total dry yield (13.40) compared to no foliar application. In summary, it was concluded that salicylic acid + sodium nitroprusside foliar application in drought stress conditions prevented the damaging effects of reactive oxygen species by strengthening the enzymatic and non-enzymatic antioxidant defense systems and prevented the peroxidation of membrane lipids by reducing the amount of malondialdehyde. The current findings show that the exogenous application of salicylic acid + sodium nitroprusside is a potential strategy for the successful cultivation of henna in drought-prone areas.
