DNA barcoding of Paracoccus marginatus (Hemiptera: Pseudococcidae) and Acerophagus papayae (Hymenoptera: Encyrtidae) in Nsukka Agro-Ecological Zone, Nigeria

The papaya mealybug Paracoccus marginatus Williams and Granara De Willink 1992 is an invasive and destructive pest of major economic importance in Nigeria. The parasitoid Acerophagus papayae Noyes and Schauff 2003 has been shown to be effective in managing P. marginatus populations. Accurate identification is essential for effective P. marginatus management and for the release of A. papayae, which is highly host-specific. This study aimed at identifying P. marginatus and A. papayea in Nigeria using DNA barcoding approach. DNA barcoding successfully identified P. marginatus and A. papayae in Nigeria with 100% similarity with the respective species in GenBank. All the P. marginatus screened were parasitized by A. papayae. This is the first report of establishment of population of A. papayae in Nigeria where the parasitoid has not previously been released. The universal barcoding primers C1J2195 and TL2N3014 initially failed to amplify P. marginatus DNA, and the challenges encountered, along with strategies for resolving them, are discussed. The establishment of population of A. papayae in the study area presents an opportunity to enhance the biological control of P. marginatus in Nigeria. This will enable encouragement, mass rearing and release of A. papayae in the region to control P. marginatus.
