Effect of cold acclimation on the supercooling point and cold hardiness substances in Heterolocha jinyinhuaphaga (Lepidoptera: Geometridae)

Heterolocha jinyinhuaphaga Chu (Lepidoptera: Geometridae) is an economically important pest on honeysuckle in China. This study investigated the physiological response mechanisms of H. jinyinhuaphaga to cold acclimation. The supercooling points (SCP) and water content of larvae, pupae, male and female moths decreased from 12 h to 84 h post cold acclimation under 5℃ and 10℃, whereas the protein, fat, and total sugar contents increased under the same temperatures as above. The SCP and water contents were lowest in larvae, male and female moths at 48 h post cold acclimation, however, they were lowest in pupae at 60 h post cold acclimation. Similarly, the protein, fat, and total sugar contents of larvae, male and female moths peaked at 48 h post cold acclimation, and they peaked in pupae at 60 h post cold acclimation. The interaction between cold acclimation temperature and cold acclimation time had non significant effect on the SCP and protein content of larvae, male and female moths (p ˃0.05), but it had a significant effect on SCP and protein content of pupae (p < 0.05) and on water, fat, and total sugar contents of larvae, pupae, male and female moths (p < 0.05). The SCP showed a positive correlation with water content of larvae, pupae, male and female moths, but showed a negative correlation with fat, protein, and total sugar contents in their bodies. This result might provide theoretical basis for future research on cold tolerance strategy and potential distribution of H. jinyinhuaphaga.
