<p><i>Sauromatum diversifolium</i>, a lesser-known member of the Araceae family, possesses tubers with significant nutraceutical value and appetite-suppressing properties. In this study, an efficient protocol for micropropagation was developed using tuber explants with apical buds. Tuber bud cultured on Murashige and Skoog (MS) medium supplemented with thidiazuron (TDZ) in combination with gibberellic acid (GA₃) exhibited high shoot induction potential. Multiple shoot regeneration response of up to 95% was recorded, with the highest shoot proliferation exceeding 50 shoots per explant, observed at 10 µM TDZ combined with 20 µM GA₃ within 60 ± 5 days of inoculation. For in vitro rooting response, indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) were tested. IAA showed superior rooting performance, with rooting percentages increasing in response to higher concentrations and up to 100% results within 50 ± 5 days of sub-culture. In contrast, high levels of IBA not only failed to induce rooting but also caused browning of the culture medium and inhibited shoot development. Acclimatization of the in vitro developed plantlets resulted in a survival rate of 65 ± 5%. These findings establish a robust and reproducible micropropagation protocol for <i>S. diversifolium</i> and open avenues for its mass propagation and nutraceutical exploration.</p>
