The impact of Bacillus subtilis NJ22, a Fe-resilient soil isolate as a potent plant Growth-Promoting agent to Spinacia oleracea L

Plant nutrition exerts significant influence on quality and quantity of agricultural productivity, the soil tends to become depleted of essential nutrients, resulting in reduced efficiency. However, soil fertilisation, and other agricultural practices are being employed to overcome such depletion and enhance soil productivity. To mitigate heavy metal pollution in soils, a promising approach is the use of PGPR (Plant Growth-Promoting Rhizobacteria). These PGPR produce phytohormones that exhibit various growth-promoting traits and can counter heavy metal toxicity. In the present study, a Fe-tolerant (up to 500 µg/mL) soil isolate, Bacillus subtilis NJ22 was isolated and characterised. The isolate showed IAA (877.75 µg/mL), Siderophore and Ammonia production, Potassium and Phosphate solubilisation and Nitrogen fixation which indicated the plant growth promoting traits of the isolate. Further, the growth-promoting ability of the strain was confirmed by observing increased root length (1.76 cm), shoot length (1.16 cm) and weight (1.5 g) of Spinacia oleracea in Fe-stressed soil. The improved plant growth in Fe-stressed soil also showed the Fe-resilience of B. subtilis NJ22 which was further confirmed by quantitative analysis of Fe in soil before and after plant growth study by Atomic Absorption Spectroscopy (AAS).
