Inhibition of advanced glycation end products formation and antioxidant activities of Psychotria luzoniensis and Psychotria banahaensis

Alzheimer’s disease (AD) is a neurodegenerative disease characterized by the presence of amyloid-beta (Aβ) plaques in the brain, and studies have linked the formation of these plaques with oxidative stress and advanced glycation end products (AGEs). Two endemic Philippine Psychotria species from Ilocos Norte, Psychotria luzoniensis (Cham. & Schltdl.) Fern.-Vill. and Psychotria banahaensis Elmer., are evaluated for their pharmacological activities. Following methanolic extraction, P. luzoniensis leaf extract (PLLE) and P. banahaensis leaf extract (PBLE) were examined for their total phenolic and flavonoid content, antioxidant activity, inhibition of AGEs formation, and anti-amyloidogenic activity. Phytochemical profiling of both Psychotria leaf extracts revealed that PLLE showed higher TPC (16.36 ± 0.87 mg GAE/g DW) and TFC (3.54 ± 0.23 mg QE/g DW) values compared to PBLE, which corresponds to its high antioxidant activity (%Decolorization = 97.33 ± 0.72% | TEAC = 183.55 ± 1.46 mg TE/kg). Regarding anti-AGE activity, despite PBLE showed a higher percent AGE inhibition, PLLE was found to be more potent (IC50 = 335.75 ppm). Interestingly, both crude extracts were able to show potential in inhibiting Aβ aggregation (percentage inhibitions of 75.23 and 65.72% at 100 μg/mL). The data indicates that both PLLE and PBLE, show promising activities that may be harnessed through further studies to formulate novel forms of treatments against AD.
