<p>The aim of this study is to qualitative assessment of β-Sitosterol (steroid), Lupeol (terpenoid), and Quercetin (flavonoid) in <i>L. reticulata</i> methanolic and ethyl acetate extracts of plant parts (leaf, stem, and root) and callus by developing HPTLC method. These extracts of <i>L. reticulata</i> plant have also been assessed for their potential antimicrobial activity on some bacterial and fungal strains. The HPTLC method is used for qualitative assessment of β-sitosterol (Steroid), Lupeol (Terpenoid), and Quercetin (Flavonoid) in <i>L. reticulata</i> methanolic and ethyl acetate extracts of plant parts (leaf, stem, and root) and callus. The antimicrobial activity of these extracts was also evaluated using the Agar well diffusion method to assess the zone of inhibition for Gram-positive bacteria <i>viz. Staphylococcus aureus and Enterococcus faecalis</i>; Gram-negative bacteria viz. <i>Escherischia coli</i>,<i> Klebsiella pneumonia</i>,<i> Pseudomonas aeruginosa</i> and fungal strain <i>Candida albicans</i>. Rf value of β-Sitosterol, Lupeol and Quercetin standard was found to be 0.55, 0.68 and 0.74 respectively. Rf value matched in all the tested plant parts and callus extracts of <i>L. reticulata</i>, indicates the presence of β-Sitosterol, Lupeol and Quercetin in all the plant parts and callus of <i>L. reticulata</i>. The antimicrobial assay results showed that the leaf, stem, and root methanolic and ethyl acetate extracts effectively inhibited all the tested bacterial strains. The callus extracts in both solvents inhibited <i>Pseudomonas aeruginosa</i>, while ethyl acetate callus extract also showed an inhibitory effect against <i>Enterococcus faecalis</i>. The tested fungal strain <i>Candida albicans</i> was effectively inhibited by all the methanolic and ethyl acetate extracts.</p>
