Lactobacillus casei modulates gut health against alzheimer’s disease: a neurodevelopmental approach

Alzheimer’s disease (AD) is a very destructive condition that which lacks viable treatments. There has been growing data indicating that the gut microbiota has a significant impact on AD. We intend to examine the potential mechanisms of the probiotic Lactobacillus casei (L. casei) via the microbiota-gut-brain axis in correcting spatial and learning memory impairments, using D-galactose (D-gal)-induced cognitive impairment in rats as the experimental model. AD has been generated by the administration of D-gal through oral route. According to the findings of this research, the performance of rats that had been treated with D-gal in Y-maze, radial arm maze and elevated plus maze test was greatly enhanced by DNP + L. casei. It has been observed elevation of superoxide dismutase (SOD) and reduction of malondialdehyde (MDA) contribute to memory formation through the L. casei treatment in combination with donepezil (DNP), which reverses the effect of D-gal. Furthermore, it was shown that the D-gal-induced dysbiosis was mitigated by enhancement of the Lactobacillus and suppression of E.coli followed by RT-PCR. In addition to this the decrease in amyloid-β levels in rats that were treated with DNP + L. casei was found beneficial. Histopathological findings displayed normal gastric epithelium, maintained villi and crypts after probiotic treatment, protecting against neurodegeneration in the hippocampus. These findings provide acceptance to the idea that L. casei could slow down the breakdown of the gut-brain-microbiota axis when combined with acetylcholine esterase inhibitor.
