Multiple gene detection of ‘Candidatus Phytoplasma australiasiaticum’ related strain associated with stem fasciation in Brassica napus L

Brassica napus L., an important oilseed crop, exhibited stem fasciation symptoms at the research farm of Sher-e-Kashmir University of Agricultural Sciences and Technology, Chatha, Jammu, in February 2025. The association of phytoplasma with symptomatic B. napus plants (variety RSPN-25) was confirmed through nested and semi-nested PCR assays using universal phytoplasma-specific primers targeting the 16 S rRNA gene (P1/Tint followed by R16F2n/R2) and the tuf gene [TUF(II)F1/TUF(II)R1 followed by TUF(II)F2/TUF(II)R1]. BLASTn analysis and phylogenetic comparisons of the amplified 16 S rRNA and tuf gene sequences revealed that the B. napus phytoplasma isolate was closely related to ‘Candidatus Phytoplasma australiasiaticum’, belonging to the 16SrII group. Furthermore, in silico RFLP analysis of the 16 S rRNA gene sequence using the iPhyClassifier tool classified the isolate within the 16SrII-D subgroup (‘Ca. P. australiasiaticum’). To the best of our knowledge, this study represents the first report worldwide of ‘Candidatus Phytoplasma australiasiaticum’ infecting Brassica napus.
