<p>The present study reports the isolation and characterization of <i>Gilmaniella humicola</i>, a rare and noteworthy fungal species, from agricultural soils of Bhauwala village in the Doon Valley, Uttarakhand, India. Preliminary identification was carried out based on cultural characteristics and morphotaxonomic features, including the color, shape, dimensions, and ornamentation of conidia and conidiophores. Molecular confirmation was achieved through amplification and sequencing of the internal transcribed spacer (ITS) region of rDNA using ITS4 and ITS5 primers, validating the identity of the isolate as <i>Gilmaniella humicola</i>. To the best of our knowledge, this represents the first documented report of <i>G. humicola</i> from the Doon Valley using an integrated morphological and molecular approach, contributing to the fungal biodiversity records of the region.</p>
