Controlled condition screening assay to simulate wilt in sugarcane using soil and crop debris inocula

The stalk-infecting vascular pathogen, Fusarium sacchari, causes wilt, a serious disease affecting sugarcane in various countries. Disease resistance is the primary management strategy and a sustainable approach in India. Currently, field screening assays for wilt resistance involve the plug method of inoculation onto standing cane stalks or planting sugarcane clones in sick-plot conditions. This process is time-consuming and resource-intensive when screening large numbers of clones. Therefore, a controlled-condition assay using infected soil and cane debris in disease-testing pits or trenches within a disease-testing chamber has been developed. In this assay, susceptible clones exhibit germination failure or post-germination shoot death, with systemic F. sacchari infections confirmed in different tissues by histological and molecular assays. The new wilt screening method is rapid and suitable for identifying wilt resistance in large numbers of sugarcane clones early in the breeding cycle. This study established a foundation for developing a quick and efficient screening assay in sugarcane.
