<p>To develop a PCR-based diagnostic method for plant DNA virus diseases, the applicability of a DNA purification-free PCR method was evaluated for detecting a notorious bipartite begomovirus, pepper yellow leaf curl virus (PepYLCV). DNA extraction was carried out by commercial kit, one-tube DNA extraction method and DNA purification-free method or homemade DNA storage paper-mediated PCR (HDP-PCR) to detect the presence of PepYLCV in field samples. Comparative analysis showed that all three methods successfully detected PepYLCV in field samples by amplifying the target 912&#xa0;bp fragment through PCR. The one-tube DNA extraction method successfully detected the virus in all tested samples, including those extracted on the sampling day and those stored at room temperature for one week. The HDP-PCR method detected the presence of pepper yellow leaf curl virus in the fresh as well as two-month-old filter paper samples, indicating the stability of viral DNA in filter paper. Further, the HDP-PCR method is the most cost- and time-effective out of the three methods used for DNA extraction.</p>
