Mass Spectrometry-Based Metabolomic Analysis and Biological Activities of Alkaloids from Haloxylon Persicum Bunge Leaves Extracts

Amaranthaceae species have garnered considerable attention due to their diverse pharmacological and biological properties. One specific species, Haloxylon persicum Bunge, is an exotic species that was accidentally introduced to the Saharan zone of Nefzaoua in southern Tunisia, North Africa. This species is known for its pharmacological and biological properties.
MethodsThis study used liquid chromatography-tandem mass spectrometry (LC-HESI-MS2) to detect alkaloid metabolites in the total alkaloids extract. The phytochemical composition (total polyphenol, flavonoid, and tannin contents) and biological activities, specifically the antioxidant and antibacterial properties of the leaf extracts of Haloxylon persicum Bunge were assessed.
ResultsThe total alkaloid extract revealed the presence of 37 chemicals, from which we identified 28 compounds, with 46.43% identified as alkaloids, including anabasine, haloxine, and taxinine. Haloxylon persicum exhibited potent antioxidant activity in multiple tests (DPPH, ABTS, TAC, and FRAP). Acid-base treatment of the hydroethanolic extract significantly enhanced its antimicrobial activity, surpassing that of the crude extract, especially, the total alkaloid and butanolic extracts.
ConclusionHaloxylon persicum and its alkaloid-derived secondary metabolites showed potential for medicinal applications and the management of pathogenic microorganisms.
