<p>This study evaluated the impact of storage duration on the polyphenolic composition, biological activities, and biodegradation potential of olive mill wastewater (OMW) derived from two Algerian olive cultivars, Zlitni and Abani. The Abani variety exhibited the highest total phenolic content, reaching (961.11 ± 65.96 µg GAE/mL) after 12 months. Qualitative analysis by liquid chromatography-mass spectrometry (LC-MS) identified 28 phenolic compounds, with kaempferol as the dominant constituent. Notably, rosmarinic acid was identified for the first time in OMW. Antioxidant activity, assessed by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), ferric reducing antioxidant power (FRAP), and hydrogen peroxide (H₂O₂) scavenging assays, showed significant variation during storage, with the Abani variety exhibiting the best IC₅₀ values at 12 months. The most potent anti-inflammatory activity, evaluated by inhibition of protein denaturation (IPD) and membrane stabilization potential (MSP), was recorded in Abani extracts at 1 month (IPD IC₅₀: 25.47 ± 1.5&#xa0;µg/mL) and 12 months (MSP IC₅₀: 16.11 ± 0.1&#xa0;µg/mL), outperforming diclofenac sodium (83.00 ± 5.75&#xa0;µg/mL and 77.66 ± 2.84&#xa0;µg/mL, respectively). Anticoagulation assays assessed using activated partial thromboplastin time (APTT) and prothrombin time (PT) revealed strong anticoagulant activity, particularly in freshly collected Zlitni OMW, with maximum values of (80.07 ± 0.15&#xa0;s) (APTT) and (37.13 ± 0.31&#xa0;s) (PT), significantly exceeding those of the negative control (30.07 ± 0.06&#xa0;s and 13.00 ± 0.1&#xa0;s). These findings confirm that storage duration markedly influences the biochemical profile and bioactivity of OMW, highlighting its potential for valorization in sustainable applications.</p>
