Virome associated with interspecific hybrid bunch grapevine cultivars in Texas

Texas is the fastest growing wine grape industry in the U.S., but little is known about the status of viruses present in vineyards across the state. To address this knowledge gap, growers and viticultural specialists were engaged to collect a total of 196 samples from 42 vineyards across 25 Texas counties from fall 2016 to summer 2017. The samples were screened by RT-PCR for 10 major viruses. Additionally, four vineyards of the interspecific hybrid bunch grape cultivar Blanc du Bois (BdB) were intensively sampled in 2021 for virome investigation by high-throughput sequencing (HTS), followed by RT-PCR screening of individual vines (n = 290) for the HTS-detected viruses. The combined results showed the occurrence of 13 distinct viruses and three viroids in Texas vineyards, including grapevine leafroll-associated virus 1 (GLRaV-1), GLRaV-2, GLRaV-3, GLRaV-4, tobacco ringspot virus, tomato ringspot virus, grapevine fleck virus, grapevine rupestris stem pitting-associated virus, grapevine virus A (GVA), GVB, GVL, GVM, grapevine red blotch virus (GRBV), hop stunt viroid, grapevine yellow speckle viroid 1 (GYSVd-1), and GYSVd-2. These graft-transmissible agents (GTAs) occurred at varying levels of incidences with ~ 50% of the vines being mixed-infected with two or more GTAs. Notably, the GTAs detected from the spatially distant BdB vineyards showed consistent monophyletic clustering patterns, suggesting their possible spread via contaminated propagative materials. The results showed that Texas’s vineyards are not immune to grapevine GTAs and reinforce the need to implement a clean grapevine program in the state.
