<p>Rice false smut, incited by an ascomycete fungus <i>Ustilaginoidea virens</i>, poses a threat to rice crops, leading to substantial economic losses in both yield and quality. The pathogen exhibits whitish yellow-green hues in culture. These colonies exhibit variable mycelial growth patterns along with circular and irregular margins. This pathogen typically infects rice filaments during the booting stage and symptoms are visible after panicle emergence. Early detection of the pathogen is imperative to implement timely management practices. Current molecular detection methods, while accurate, are time-consuming, necessitate sophisticated equipment, and require skilled personnel. Thus, a rapid, highly specific detection assay is warranted. Targeting <i>UvG- β1</i>gene of <i>U. virens,</i> using a set of four LAMP primers, enabled the detection of the pathogen within a mere 50&#xa0;minutes at a temperature of 60&#xa0;°C with colorimetric visualization of LAMP products. In this study, we have successfully standardized a Loop-Mediated Isothermal Amplification (LAMP) assay for the specific detection of <i>U. virens</i>. The developed LAMP assay proves highly specific and sensitive, capable of detecting as little as 100 femtograms (fg) of template DNA. The assay’s reliability and accuracy have been validated through testing with thirty study isolates and twenty-two field samples collected from boot leaf sheaths. Overall, the LAMP assay represents an affordable, specific, and efficient solution for the early detection of the rice false smut pathogen, enabling intervention measures before visible symptoms manifest and holds great promise for improving the management practices and safeguarding rice crops against substantial losses.</p>
