Objective <p>This study aimed to evaluate and compare the antimicrobial efficacy of a benzoylthiourea derivative (HL), methylene blue (MB), diode laser applications at different wavelengths, and conventional disinfectants (NaOCl, DMSO, NaCl) against E. <i>faecalis</i> biofilm in root canals, with a particular focus on the photosensitizing potential of HL in photodynamic therapy.</p> Materials and methods <p>Eighty-two single-canaled lower premolar teeth were inoculated with <i>E. faecalis</i> and incubated for 22 days. Two samples were analyzed using SEM to confirm biofilm control. Outliers and contaminated samples were excluded (16 samples) to ensure an equal distribution among the groups. The remaining 64 teeth were randomly divided into 8 groups (<i>n</i> = 8) according to the decontamination protocol: G1: 5% NaOCl (positive control) for 5&#xa0;min, G2: DL; 976-nm diode laser (1&#xa0;W, 3 cycles of 20&#xa0;s each) G3: MB/DL; Methylene blue (100 µL) / 650-nm DL (200 mW, 60&#xa0;s) conventional photodynamic therapy (PDT), G4: HL/ DL; HL (100 µL) / 450-nm DL (200 mW, 60&#xa0;s), G5: Dimethyl sulfoxide (DMSO) for 5&#xa0;min, G6: MB; MB (100 µL) for 5&#xa0;min, G7: HL; HL for 5&#xa0;min, G8: NaCl; NaCl for 5&#xa0;min. The antibacterial activity was evaluated by counting the colony-forming units based on the reduction in colony numbers.</p> Results <p>The greatest reduction in colony count (99.1%) was observed in the NaOCl group. However, no significant differences were observed in colony reduction between the NaOCl group and the DL (97.3%), HL (96.1%), and HL/DL (92.4%) groups (<i>P</i> &gt; 0.05). The results from the NaOCl, DL, HL, and HL/DL groups demonstrated a significantly higher reduction in colony count compared to all other groups. The comparison between the NaCl (42.7%) and the MB (64.4%), MB/DL (43.7%), and DMSO (79.6%) groups showed comparable results with no statistically significant variation (<i>P</i> &gt; 0.05).</p> Conclusion <p>The results suggest that photodynamic therapy modified with HL, as well as HL alone, has the potential to eliminate <i>E. faecalis</i> from root canals. Further studies are required to assess their effectiveness and clinical applicability.</p>

Objective

Results
