Purpose <p>This study set up to evaluate the efficacy of diode laser conjugated 2% Chlorhexidine&#xa0;digluconate (CHX) and 2.5% sodium hypochlorite (NaOCl) in removing <i>Enterococcus faecalis</i> (E. <i>faecalis</i>) from the root canals.</p> Methods <p>Ninety extracted single rooted, single canal teeth with intact mandibular bone were provided, the temporary filling material was removed by washing with normal saline, ethylenediaminetetraacetic acid (17%), and sodium hypochlorite (2.5%), and the roots were sterilized for 20&#xa0;min at 120&#xa0;°C. All samples were contaminated by suspension of E. <i>faecalis</i> for four weeks, all of specimens were randomly categorized into six groups (<i>n</i> = 15 in each group)<Emphasis Type="Underline">.</Emphasis> After decontamination treatments (2.5% NaOCl, 2% CHX, Normal saline, 2.5% NaOCl + Diode laser, 2% CHX + Diode laser, Normal saline + Diode laser), The diode laser was done at 980&#xa0;nm, 1.5W output for 5 × 15&#xa0;s. samples collection was done and specimens transferred onto blood agar culture medium. The Kruskal–Wallis test was used to compare the CFU values of the bacteria and the level of significance was set at 0.05.</p> Results <p>The suppression of bacterial proliferation in all the groups was notable compared to the control group (<i>P</i> &lt; 0.05). The effect of 2.5% NaOCl accompanied with laser diode was significantly better than that of the 2.5% NaOCl (<i>P</i> &lt; 0.001) and in CHX 2% + Diode laser group the results were similar.</p> Conclusion <p>Based on results, 2% CHX, 2.5% NaOCl associated with diode laser achieved success by reducing root canal infection, so diode laser can boost common root canal disinfection protocols applying 2.5% NaOCl and 2% CHX.</p>
