<p>This study assessed fecal contamination across seawater, interstitial water, and sediment microhabitat at Olho D’Água Beach, São Luís, Brazil, comparing results to Brazilian bathing standards. Samples were obtained from Olho D’Água Beach, located in São Luís, Maranhão, comprising three samples collected during the dry season and three during the rainy season. Qualitative and quantitative analyses of total coliforms, <i>Escherichia coli</i>, and <i>Enterococcus</i> spp. were carried out using the chromogenic enzyme substrate method. The highest average microorganism concentrations were recorded during the rainy season. Among microhabitats, total coliforms showed similar high densities in surface water and interstitial water (2.4 × 10<sup>4</sup> MPN/100 mL<sup>-1</sup>), followed by sediment (1.6 × 10<sup>4</sup> MPN/100 mL<sup>-1</sup>). <i>E. coli</i> and <i>E.</i> spp. exhibited their highest average densities in surface water (5.5 × 10<sup>3</sup> and 8.8 × 10<sup>3</sup> MPN/100 mL<sup>-1</sup>, respectively), with lower concentrations in interstitial water and sediments. Although no significant differences in microbial densities were detected among microhabitats overall, a significant difference was observed during the rainy season. Microorganism levels exceeding Brazilian regulatory guidelines were detected in all microhabitats. These findings highlight the importance of considering different microhabitats when monitoring beach environmental quality. Understanding spatial and seasonal variations in fecal contamination is essential for improving coastal management strategies and safeguarding public health in tropical urban beaches.</p>
