<p>An intensive investigation was conducted to assess the stock status of four commercially important portunid crabs viz.,<i> Portunus pelagicus</i>,<i> P. sanguinolentus</i>,<i> P. gladiator</i>, and <i>Charybdis natator</i> along the Gulf of Mannar, southeast coast of India, from October 2021 to September 2022. Weekly data from four landing centers (Vedalai, Keelakarai, Therespuram, and Thiruchendur) was analyzed using Catch per Unit Effort (CPUE). The bottom set gillnet, trammel net, and trawl net were the primary gears used. Therespuram recorded the highest CPUE for all the studied species using bottom set gillnets, with <i>P. pelagicus</i> (11.51&#xa0;kg/day) and <i>C. natator</i> (9.58&#xa0;kg/day) peaking in February and June, respectively. <i>P. sanguinolentus</i> (3.08&#xa0;kg/day) and <i>P. gladiator</i> (2.28&#xa0;kg/day) peaked in April and August. With trammel net operated on the coast of Vedalai and Keelakarai, the maximum value (2.28Kg/day) was observed for <i>P. pelagicus</i> in June (1.84 Kg/day); for <i>P. sanguinolentus</i> the same was recorded in the month of March (1.97 Kg/day) at Vedalai. For the trawl net, the CPUE was found to be maximum during October for all studied species except for the species <i>P. gladiator</i> which shows maximum value in February. Annual catches recorded for the species <i>P. pelagicus</i>, <i>P. sanguinolentus</i>, <i>P. gladiator</i> and <i>C. natator</i> were 147.74303 tonnes, 46.56905 tonnes, 24.65543 tonnes and 83.00259 tonnes respectively. Maximum Sustainable Yield (MSY) estimates were highest for <i>P. pelagicus</i> (137.8324 tonnes), followed by <i>C. natator</i> (109.0144 tonnes). Annual catches exceeded MSY for <i>P. pelagicus</i> and <i>P. sanguinolentus</i> indicating optimal exploitation, whereas <i>C. natator</i> was underexploited. Exploitation ratio were found to be 0.54, 0.57, 0.51 and 0.38 for <i>P. pelagicus</i>, <i>P. sanguinolentus</i>, <i>P. gladiator</i> and <i>C. natator</i> respectively. The study suggests prioritizing bottom set gillnets for sustainable crab fishing and implementing management strategies for conservation.</p>
