<p><i>Vibrio parahaemolyticus</i> is a pathogen of significant importance in the seafood sector, and the increased incidence of antimicrobial resistance in this pathogen poses a serious threat to seafood safety. To investigate this issue, a total of 90 samples comprising 78 fishes (46 finfish and 32 shellfish) and 12 water samples were collected from two fish landing centres in Thoothukudi, Tamil Nadu, India: the motorized boat landing centre at Therespuram (LC1) and the mechanized boat landing centre at the Fishing Harbour (LC2). A total of 126 suspected <i>V. parahaemolyticus</i> colonies were isolated among which&#xa0;biochemical testing identified 37 isolates (29%) as presumptive <i>V. parahaemolyticus</i>. Among them, 29 (78.4%) were positive for <i>toxR</i> gene PCR confirmation, while only 8 exhibited the Kanagawa phenomenon. The shrimp samples showed a higher prevalence of <i>V. parahaemolyticus</i> (10%) than the crabs (6.6%), finfishes (5.5%), and water (1.1%) samples. The prevalence of virulence factors in <i>V. parahaemolyticus</i> are as follows: <i>tdh</i> 2 (7%), <i>trh</i> 7 (24%), <i>tlh</i> 29 (100%) and <i>T3SS1</i> genes 25 (86%). Antimicrobial susceptibility testing (AST) was done using 11 antibiotics belongs to 10 different classes. 93% of the isolates were resistant to trimethoprim, and 14% and 7% of isolates were resistant to cefoxitin and ceftazidime respectively. Higher Multiple Antibiotic Resistant (MAR) index values (&gt; 0.2) were observed in 28% of the isolates. This study underscores the critical role of routine monitoring, strict hygiene practices, and correct seafood handling procedures in maintaining food safety.</p>
