Therapeutic Potential of Microalga Scenedesmus vacuolatus: In Vitro Evaluation of Antioxidant and Anticancer Activities

The present study reports the stress-induced secondary carotenogenesis in the nutraceutically valuable Chlorophycean microalga Scenedesmus vacuolatus isolated from the Vembanad estuary, Kerala. The isolate was tested in five different algal culture media to analyze the growth rate and a two-stage cultivation system was adopted to produce carotenoids. Under nitrogen-limited conditions, the reddish aplanospore produced secondary carotenoids. The carotenoids were analyzed using High-Performance Liquid Chromatography. As the major carotenoids, S. vacuolatus accumulated canthaxanthin, β-carotene, astaxanthin, and lutein. Free radical scavenging properties of carotenoids were assessed through a DPPH assay. In vitro, cytotoxic activity against the human cervical cancer (HeLa) and human colon cancer (HT-29) cell lines was investigated. Cell viability was significantly reduced to 53.56% against HeLa cell lines and 49.92% against HT-29 cell lines. The results suggested that the microalga S. vacuolatus is a promising source for producing carotenoid-rich biomass. Furthermore, it also exhibited significant antioxidant and anticancerous activities indicating its nutraceutical significance.
