<p>The strawberry tree (<i>Arbutus unedo</i> L.) has been used in traditional natural healing practices for centuries, particularly in Mediterranean regions. This work investigates the bioactive potential of <i>A. unedo</i> leaves collected from the ‘Guelma’ region (Algeria). Phytochemical screening confirmed that the aqueous extract contained high levels of polyphenols, flavonoids, and proanthocyanidins, demonstrating significant antioxidant activity (DPPH IC<sub>50</sub> = 95.4&#xa0;μg/mL; total antioxidant activity: 85.11 ± 0.09&#xa0;mg GAE/g DE). The extract exhibited potent antibacterial effects, particularly against Gram-positive bacteria, and inhibited mushroom tyrosinase, suggesting dermatological applications. Interestingly, it displayed anti-inflammatory properties by suppressing pro-inflammatory nitric oxide (NO) production (IC<sub>50</sub> = 60.87 ± 0.33&#xa0;µg/mL). Cytotoxicity assays revealed no adverse effects on RAW 264.7 macrophage cells (LC₅₀ = 732.88 ± 1.5&#xa0;µg/mL), with a favorable selectivity index (SI = 12). Through TLC-bioautography and GC–MS analysis, arbutin was identified as the primary active compound. Given these findings, the aqueous leaf extract of <i>A. unedo</i> shows promise as a natural preservative for the food industry, as well as a functional ingredient for pharmaceutical and cosmetic formulations.</p>
