Curcumin Delivery in Regenerative Medicine for Alzheimer’s Disease

Curcumin has been widely used as a therapeutic and dietary agent. It possesses several beneficial properties, including the antioxidant, anti-inflammatory, regenerative, anti-protozoal, anti-bacterial, anti-apoptotic, and anti-tumor activities. Notably, curcumin exhibits potential in inhibiting amyloid-β-protein (Aβ) aggregation. Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by cognitive decline, behavior changes, and Aβ aggregation. However, as a lipid-soluble agent, curcumin has limited ability to cross the blood-brain barrier and reach brain tissue.
MethodsIn this study, we conducted a literature review on curcumin carriers in regenerative medicine aiming at treating of Alzheimer’s disease.
ResultsWe reviewed curcumin properties and its effects on Alzheimer’s disease in both in vitro and in vivo studies, including the molecular mechanisms involved. Various delivery strategies for curcumin in the context of Alzheimer’s disease were discussed, focusing on nanostructures and tissue-engineered scaffolds, particularly injectable hydrogels and self-assembled scaffolds, as carriers. Furthermore, alternative routes of administration and stem cell-based therapies as a new approach for the treatment of Alzheimer’s disease were discussed. Finally, clinical trials of curcumin for Alzheimer’s disease were reviewed.
ConclusionMultiple strategies in regenerative medicine, including tissue engineering scaffolds (such as injectable hydrogels and self-assembled nanofibers), and nano-constructs in nanotechnology (liposomes, micelles, nanogels, polymeric nanoparticles, solid-lipid nanoparticles, and dendrimers) have been developed to enhance curcumin delivery and overcome the blood-brain barrier for Alzheimer’s disease treatment.
