Comparative analysis of methodologies for disease aggressiveness evaluation of Phytophthora capsici in pepper plants

The aim of this study was to evaluate the pepper (Capsicum annuum) response to six strains of Phytophthora capsici. Three inoculation methodologies were evaluated, using (i) toothpicks, (ii) infested rice grains and (iii) culture medium discs containing the pathogen. The applicability of these methods was explained through the analysis of severity and area under the disease progress curve (AUDPC) in plants of the California Wonder variety infected with six strains of P. capsici collected in the Manabí province, Ecuador. P. capsici isolates showed variations in their morphological and cultural characteristics, as well as significant differences (P ≤ 0.05) in the levels of aggressiveness and inoculation methodologies. All plants inoculated with the six P. capsici strains displayed rot symptoms of root, crown and stem, as well as wilt at different intensities. Higher levels of severity and AUDPC were achieved in plants inoculated with infected rice grains, whereas the toothpick methodology offered a better fit of the linear regression slope over the evaluation time. Among the three methodologies correlated using the different parameters of the disease, only significant high correlation was observed between AUDPC of infested rice grains and severity (P = 0.0271), AUDPC (P = 0.0269) and slope (P = 0.0382) of wooden toothpick. Based on the results, the use of the infested rice grains and toothpick methodologies could represent important alternatives for the artificial inoculation of P. capsici in Capsicum breeding research, considering their practicality as well as lower variation in the standard deviation of the samples.
