Purpose of Review <p>Breast cancer's heterogeneity poses significant treatment challenges, driving exploration of plant-based therapies. <i>Clinacanthus nutans</i> (<i>C. nutans</i>), a well-known perennial plant rich in bioactive compounds, has gained recognition for its potential against breast cancer. However, existing studies mostly investigate plant extracts without detailed knowledge of specific bioactive compounds, limiting targeted therapy development. This paper reviews prominent compounds within <i>C. nutans</i> extracts, with a focus on β-sitosterol, stigmasterol, shaftoside, and lupeol, known for their cytotoxic effects against breast cancer.</p> Recent Findings <p>Our review reveals isolated compounds’ significant apoptotic-inducing potential against MCF-7 and MDA-MB-231 breast cancer cells, involving mechanisms such as cell cycle arrest, mitochondrial disruption, apoptotic gene regulation, caspase activation, and reactive oxygen species (ROS) scavenging. We also documented the combination effects of β-sitosterol, stigmasterol and lupeol with current chemotherapeutics, demonstrating good synergism with tamoxifen, sorafenib, doxorubicin and 5-fluorouracil.</p> Summary <p>This review serves a dual purpose: firstly, to substantiate the scientific basis for the claims associated with <i>C. nutans</i> and its market products, and secondly, to introduce potential bioactive compounds of <i>C. nutans</i> origin that could pave the way for innovative drug development. In the ongoing battle against breast cancer, <i>C. nutans</i> and its bioactive constituents offer promising avenues for exploration and advancement.</p> Graphical Abstract <p></p>
