Comprehensive Proteomic Profiling and Functional Network Analyses of Indigenous Zebu (Bos indicus) Cattle Spermatozoa

Indigenous zebu cattle (Bos indicus) are known for their uniqueness in terms of high fertility even under a low-input production system. Investigating sperm proteomic profile would help in a better understanding of unique sperm cellular and molecular mechanisms in zebu cattle. Therefore, the current study was undertaken to characterize and quantify the indigenous zebu (Deoni breed) bull sperm proteome using a high throughput mass spectrometry technique. We identified 1441 proteins in spermatozoa; 12% (n = 173) of proteins were observed to contain signal peptides indicating their secretory nature. The most abundant proteins in zebu bull spermatozoa included A-kinase anchoring protein 4, Seminal plasma protein A3, Cationic trypsin, C-type natriuretic peptide, Outer dense fibre protein 2, Spermadhesin-1, Seminal ribonuclease, Seminal plasma protein PDC-109, Metalloproteinase inhibitor 2 and C–C motif chemokine 2; all these proteins have a prominent role in fertility. Gene ontology analysis of identified proteins revealed that most proteins were involved in spermatogenesis, sperm motility, capacitation, sperm-egg recognition and sperm binding to zona pellucida. The proteins significantly enriched in spermatozoa were found to be involved in the metabolic pathway and Oxidative phosphorylation. Collectively, the study provides a comprehensive description of the zebu bull sperm proteome and their functional association, which could be used as a reference for further studies on the fertility and protecting the genetic diversity of zebu cattle.
