<p>Cisplatin-related toxic side effects, including muscle atrophy, body weight loss, and anorexia, significantly impair the quality of life in cancer patients. Herbal medicines with antioxidant and anti-inflammatory properties have shown potential to mitigate cisplatin-induced side effects. <i>Humulus scandens</i> (HS), a traditional medicinal plant with strong antioxidant and anti-inflammatory activities, has not yet been investigated for protective effects against cisplatin-induced toxicity. Therefore, we conducted a study to evaluate the effects of HS extract administration on alleviating cisplatin-induced adverse effects in mice. Eighteen healthy male C57BL/6 mice were used in a 35-day experiment. Animals were randomly assigned to three groups: (1) control group, which received only regular drinking water; (2) cisplatin-treated group, which administered with cisplatin (3&#xa0;mg/kg body weight, intraperitoneally) daily from days 28 to 35 while receiving regular water; and (3) Cis plus HS group, which received the same cisplatin treatment from days 28 to 35 and drinking water supplemented with HS extract (750&#xa0;mg/kg). The results demonstrated that cisplatin treatment caused marked inflammation and oxidative stress in skeletal muscle tissues. It also decreased appetite, induced muscle atrophy, reduced body weight, and impaired muscle strength. In contrast, HS extract administration partially alleviated these negative effects. Specifically, HS extract administration reduced inflammation and oxidative stress in muscle tissues, attenuated muscle atrophy, and improved muscle strength in mice. In conclusion, these findings suggest that HS extract may serve as a beneficial dietary supplement to alleviate cisplatin-induced muscle atrophy.</p>
