Optimization of [6]-gingerol extraction methods from ginger rhizomes for enhanced bioactivity

This study aimed to optimize extraction methods for maximizing [6]-gingerol production while minimizing its degradation. Various drying methods, extraction solvents, extraction methods, extraction conditions such as temperature and time, and biological activities were examined. Freeze-dried ginger exhibited significantly higher [6]-gingerol content compared to ginger subjected to heat treatment. Among the extraction solvents, hexane showed the highest efficiency in extracting [6]-gingerol. [6]-Gingerol were maximally extracted in the reflux extraction. To further enhance extraction efficiency for both reflux and ultrasonic methods, a response surface methodology (RSM) was employed. The results showed that [6]-gingerol was maximally extracted within 31.9 min at 62.5–64.4 °C for ultrasonic extraction and within 35.8 min at 66–78.6 °C for reflux extraction. Correlation analysis revealed that [6]-gingerol showed a similar correlations with DPPH radical scavenging activity and ABTS radical scavenging activity compared to [6]-shogaol. Additionally, [6]-gingerol demonstrated stronger correlations with anti-inflammatory and anti-cancer activities compared to [6]-shogaol. To the best of our knowledge, this endeavor heralds a pioneering examination of the intricate interplay between various drying methodologies and bioactivities (anti-cancer and anti-inflammatory activity), synergistically aligned with RSM optimization.
