<p>Trichome, uni- or multicellular structures that look like fine hairs is a modified epidermal cell(s) of a plant and has various forms. Trichomes act as a physical barrier, protecting the plant from herbivores (i.e. insects) and pathogens (i.e. fungi) and also secrete various defensive secondary metabolites. In this study, we aimed to understand molecular details underpinning trichome production in radish. For this purpose, we performed transcriptomic analysis between a trichome-poor ‘Jinjudaepyung (JD)’ and a trichome-abundant ‘Simu (SM)’ inbred line. Five genes among 65 radish trichome-related genes were differentially expressed between two lines. Especially, radish <i>MYB23</i> homolog (<i>RsMYB23</i>, Rs189000) involved in the trichome initiation was down-regulated in the trichome-poor JD line compared to the trichome-abundant SM line. In addition, overexpression of <i>RsMYB23</i> in <i>Arabidopsis</i> model plant resulted in the higher abundance of trichomes than that of wild type plant. Our study showed that <i>RsMYB23</i> is required for the trichome initiation in the radish, expanding our understanding on trichome development in the radish plant.</p>
