Relevant literature was gathered from PubMed and Google Scholar, focusing on studies examining Klotho levels, gene polymorphisms, and therapeutic implications. Keywords included “Klotho,” “diabetes,” “diabetic complications,” “diabetic nephropathy,” “ diabetic neuropathy,” “diabetic retinopathy,” “diabetic cardiovascular complications,” “diabetic liver disease,” and “diabetic foot syndrome.”</p> Results <p>Evidence implied that reduced α-Klotho is associated with diabetes progression and its complications, including nephropathy, retinopathy, neuropathy, and cardiovascular diseases. Some polymorphisms of the Klotho gene influence susceptibility to diabetes and its complications. The identification of Klotho gene polymorphisms provides valuable insights into personalized medicine approaches for managing diabetes. The presence of conflicting reports regarding Klotho levels in diabetic patients highlights the need for standardized assessment methods. Further research is needed to establish Klotho measurement techniques, explore its therapeutic potential, and conduct large-scale clinical trials to validate its efficacy in managing diabetic complications.</p> Conclusion <p>Klotho is emerging as a promising biomarker and therapeutic target in DM.
