Master Orchestrators of Cellular Regulation: Advances in Protein Kinase Inhibitors for Therapeutic Intervention

Protein kinases, owing to their pivotal role in regulating diverse cellular functions, stand out as a crucial enzyme class for uncovering therapeutic agents with significant implications for human disease. Given their capability to add phosphate residues to target proteins, protein kinases essentially function as master orchestrators governing nearly all cellular processes. These biologically captivating catalysts have garnered an expanding knowledge base, gaining recognition for their role in cellular regulatory control and their increasing significance in successful therapeutic intervention. The exploration and clinical advancement of protein kinase inhibitors, designed to target a spectrum of diseases, are advancing steadily. Numerous protein kinase inhibitors are currently undergoing clinical trials, and ongoing preclinical research is actively identifying new chemotypes that bind to distinct regions in the protein, along with the development of isoform-selective compounds. The pursuit of novel protein kinase inhibitor scaffolds persists, particularly those that can serve as platforms for modification to modulate selectivity profiles. This review aims to spotlight advancements in the development of protein kinase inhibitors.
