<p>Bovine mastitis and reproductive disorders present significant challenges to dairy farming, leading to substantial economic losses globally. In this study, genetic profiling of a targeted region of the TLR4 gene was conducted in Hardhenu cattle. Using PCR–RFLP, a SNP at the g.107062971A&gt;B loci was genotyped. Only two genotypes (AA and AB) were identified in the targeted population, with the A allele being more prevalent. Chi-square and logistic regression analyses revealed significant associations between these genotypes and clinical mastitis and reproductive disorders. Animals with the AB genotype showed significantly reduced susceptibility to clinical mastitis (3.27 times) and reproductive disorders (8.50 times) compared to those with the AA genotype (<i>P</i> &lt; 0.05). However, least squares regression analysis did not find substantial relationships between genotypes and key performance traits (total milk yield, 305-day milk yield, lactation length, and peak yield; <i>P</i> &gt; 0.05). These findings suggest that incorporating the identified TLR4 gene variants into selection criteria may enhance disease resistance without affecting milk production traits. Nonetheless, further validation with a larger sample size is essential to strengthen these associations and facilitate their practical implementation.</p>
