Simultaneous determination of patulin, 5-hydroxymethylfurfural, acrylamide, and Ochratoxin A in coffee, tea, and soy sauce by HPLC-DAD

Mycotoxins are toxic secondary metabolites produced by various species of fungi. They are natural contaminants of agricultural crops, such as cereals, fruits and coffee beans. Consumption of mycotoxin-contaminated food can cause acute toxicity in humans and animals. Acrylamide (ACM) and 5-hydroxymethylfurfural (HMF) are heat-induced food toxicants produced during the Maillard reaction. Both ACM and HMF are potentially carcinogenic to humans. This study reports, for the first time, a simple and fast HPLC-DAD method for the simultaneous determination of two mycotoxins (ochratoxin A, OTA, and patulin, PAT) and two Maillard reaction compounds (ACM and HMF) in foods. For solid food samples, the targeted compounds were extracted using a solvent mixture of methanol (MeOH) and 0.1% CH3COOH (50:50 v/v) with ultrasonication followed by vortexing. For liquid food samples, they were diluted 1:100 with the same mixture and vortexed for 2 min prior to HPLC analysis. Under optimized chromatographic conditions, separation using gradient elution coupled with a C18 column provided the best sensitivity and selectivity. The satisfactory recoveries and high selectivity of the developed method demonstrate its great potential practical applications.
